The Cognitive Stack: Lion's Mane, L-Theanine and NMN — and Why Each Targets a Different Layer of Brain Performance

Most people who take nootropics pick one compound and wonder why it is not enough. The answer is that cognitive performance has multiple failure modes, each with a different mechanism. A complete protocol addresses all of them simultaneously — and the compounds do not overlap.

The word "nootropic" has been applied to everything from caffeine to racetams to lion's mane mushroom, which makes it almost useless as a category. What it obscures is that different cognitive compounds work through entirely different mechanisms — and that means taking multiple compounds is not redundant. It is necessary, if the goal is to address cognitive performance comprehensively rather than to nudge one variable.

The cognitive stack we use and recommend at Alchemy Fit addresses three distinct layers of brain function: structural neuronal maintenance, acute neurotransmitter environment, and cellular energy supply. Here is what that means in practice.

Layer 1Structural — Lion's Mane & NGF
Layer 2Environment — L-Theanine & alpha waves
Layer 3Energy — NMN & NAD+

Layer 1: Structural — Lion's Mane and NGF

Lion's Mane (20:1 extract) operates at the structural level. Its active compounds — hericenones and erinacines — stimulate the production of Nerve Growth Factor, the protein responsible for the growth, maintenance and survival of neurons. NGF is not a neurotransmitter. It does not produce a transient chemical shift. It supports the physical substrate on which all cognitive function depends.

NGF levels decline with age. The cognitive deterioration most people experience after 35 — reduced processing speed, memory fragility, reduced neuroplasticity — is at least partly a consequence of this decline. Lion's Mane directly addresses it. The effect is cumulative and requires consistent supplementation over weeks to months before it is fully established. This is not a compound you feel on day one. It is the foundation layer.

Take 500mg daily, consistently. The research protocols showing meaningful cognitive outcomes ran for sixteen weeks. Read our full Lion's Mane explainer here.

Layer 2: Environment — L-Theanine and Alpha Wave Activity

L-Theanine (600mg) operates at the neurotransmitter environment level. It promotes alpha wave brain activity — the electrical pattern associated with relaxed alertness, present when you are calm, focused and not under acute stress. It modulates GABA, reduces glutamate, and produces a calming effect without sedation.

The distinction from the other layers is that this is the layer that addresses interference. Most people's problem with focus is not that their brain lacks structural capacity — it is that the internal noise level (stress, cortisol, background anxiety) prevents them from accessing the capacity they have. L-Theanine at 600mg clears that interference. Read our full L-Theanine explainer here.

Layer 3: Energy — NMN and NAD+

The brain is the most metabolically demanding organ in the body. It consumes approximately 20% of total energy despite representing roughly 2% of body mass. That energy is produced by mitochondria, and mitochondrial efficiency depends on NAD+. As NAD+ declines with age — starting in the late twenties — the brain's energy supply becomes increasingly limited.

This is the layer that underlies the other two. A structurally intact brain running on insufficient cellular energy will still underperform. NMN 500mg restores the NAD+ precursor supply. Stack it with TMG 600mg to protect the methylation pathway. Read the full NAD+ explainer here.

"Structure, environment, energy. Three layers. Three compounds. None of them redundant. The person who says 'I tried Lion's Mane and felt nothing' usually stopped at layer one and never addressed the layer underneath it."

Alchemy Fit — The Science of Protocol

The Full Protocol

Morning with water or a light meal: NMN 500mg + TMG 600mg. These work throughout the day on the cellular energy layer and methylation support.

Lion's Mane 500mg daily — timing is flexible. Some prefer morning for a consistent routine; some prefer with lunch. Consistency matters more than timing for a compound operating on the structural layer.

L-Theanine 600mg as needed — most effectively used in the morning before cognitively demanding work, or in the evening when cortisol load is high and the goal is decompression before sleep. For sustained cognitive clarity without stimulants, daily morning dosing is the recommended approach.

Run the full stack for a minimum of ninety days. Layer 2 (L-Theanine) you will feel within days. Layer 3 (NMN) typically makes itself known at four to six weeks. Layer 1 (Lion's Mane) builds over the full ninety-day period. Why ninety days is the correct assessment window — read here.
