The Complete Longevity Stack: NMN, TMG, R-ALA and Lion's Mane — What Each Does and Why All Four Are Necessary

Longevity supplementation is not about a single compound. It is about addressing the multiple, interacting mechanisms through which biological ageing occurs. Four compounds. Four distinct mechanisms. One protocol designed around how they work together rather than how they look on a label.

The longevity supplement space has a marketing problem. Every compound is presented as the answer — the one molecule that will reverse ageing, restore energy, and add decades. This is effective advertising and poor science. Biological ageing is not a single mechanism. It is the accumulation of damage across multiple systems simultaneously. Compound 1: NMN — The Fuel Supply

Nicotinamide Mononucleotide (NMN) is an immediate precursor to NAD+ — the coenzyme that drives mitochondrial energy production, fuels the sirtuin longevity proteins, and enables the PARP enzymes responsible for DNA repair. NAD+ levels decline by approximately 50% between age 20 and 50. NMN restores the precursor supply. Human clinical trials have consistently demonstrated that NMN supplementation raises NAD+ levels in blood and tissue. This is the foundation compound. The protocol does not work without it.

Compound 2: TMG — The Pathway Protector

Trimethylglycine (TMG) is a methyl donor that protects the methylation cycle from the depletion that sustained NMN supplementation causes. NMN raises NAD+ through a pathway that consumes methyl groups as a byproduct. Without adequate methyl donors, this depletion manifests as elevated homocysteine, slightly impaired mood and cognition, and reduced efficiency in the pathways that NMN is supposed to be supporting. Compound 3: R-ALA — The Oxidative Defence

R-Alpha Lipoic Acid addresses the oxidative damage that mitochondrial energy production generates as a byproduct. Raising NAD+ increases mitochondrial activity — which means more ATP and also more reactive oxygen species. Without antioxidant support, this increased activity accelerates oxidative damage to mitochondrial membranes and DNA. R-ALA neutralises ROS within the mitochondria themselves (one of very few antioxidants capable of this), and regenerates vitamin C, vitamin E and glutathione back to their active forms — extending the protective capacity of the body's entire antioxidant network. Compound 4: Lion's Mane — The Structural Layer

Lion's Mane (20:1 extract) is the cognitive and neurological dimension of the longevity protocol. Its hericenones and erinacines stimulate Nerve Growth Factor production — supporting the growth, maintenance and survival of neurons at the structural level. NGF declines with age, and the cognitive deterioration associated with ageing is partly a consequence of reduced NGF activity and neuroplasticity. Dose: 500mg daily. Takes the longest of the four compounds to establish — allow sixteen weeks for full assessment.

How to Run the Protocol

Morning, with water or a light meal: NMN 500mg + TMG 600mg + R-ALA 600mg. These three are taken together — morning dosing aligns with the circadian rhythm of NAD+ metabolism, and the antioxidant support is most useful when mitochondrial activity is highest during waking hours.

Lion's Mane 500mg: morning or midday, on a consistent daily schedule. Consistency matters more than the specific time for a compound operating on the structural NGF pathway.

Run for a minimum of ninety days before assessing. The protocol compounds on itself — the longer it is maintained, the more stable the NAD+ baseline, the more established the NGF stimulus, the more protected the methylation cycle.
